Assessing leaf redox status in plants using reduction-oxidation Green Fluorescent Proteins (roGFP1)

Ratio imaging is a very useful tool in live cell microscopy to carry out both quantitative and qualitative measurements. Besides, ratiometric measurements have the advantage of being independent of the concentration of the marker.

In this seminar, we present a set of experiments in order to analyze the cellular redox state of Arabidopsis thaliana by ratio imaging with the reduction-oxidation sensitive green fluorescent protein (roGFP1) gene. The expression of this gene is targeted either at the cytosol or at the mitochondria of the plants.

Different approaches have been tried to perform these analysis, mainly confocal and fluorescence microscopy. Finally a conventional fluorescent microscope with proper filters was used . In terms of image analysis, two different softwares have been also tried to finally perform an automatic analysis using ImageJ (an open source software).
