In this workshop, we will be able to discover the Biolistics Gene Gun technology and its applications through the new Helios Gene Gun from Bio-Rad.

Biolistics is a particle delivery system based on a helium pumped gun. It is a direct physical method for transfection or labelling. The microparticles of gold or tungsten coated with DNA, siRNAs or lipophilic dyes are shot against the biological sample penetrating cellular membranes and cells. This method is suitable for many types of tissues, organs and cells, in in vitro or in vivo experiments; but it also allows the labelling of previously fixed tissue or organ.

This technique has different applications:
• Labelling of fixed neurons in sections or organotypic cultures of hippocampus, striatum, cortex… allowing visualization of the axonal and dendritic network and the study of spinogenesis changes and spine morphology.

• As a cellular transfection method is highly efficient (up to >90%), also in the case of multiple genes transfections. It is specially indicated in transfection resistant cells such as neuronal or primary cell cultures. In differentiation processes it allows to transfect cells at the desired differentiation stage.

• Transfection method in live animals, embryos or organotypic cultures. Main in vivo applications are in brain, tumours, liver, spleen, kidney, pancreas
